Assessment of the cytolytic potential of a multivirus-targeted T cell therapy using a vital dye-based, flow cytometric assay

This paper evaluates a flow cytometric cytotoxicity assay for multiVSTs. It highlights that multiVSTs, expanded in G-Rex 5M devices for 14 days, show potent cytolytic activity against viral targets that is better detected by this new assay than by chromium release.

Inosine Induces Stemness Features in CAR-T cells and Enhances Potency

The study shows that inosine induces stemness and metabolic reprogramming in CAR-T cells, improving potency. Clinical-scale manufacturing of GD2-CAR-T cells in inosine-containing media was performed using a semi-closed G-Rex system.

Antiviral cellular therapy for enhancing T-cell reconstitution before or after hematopoietic stem cell transplantation (ACES): a two-arm, open label phase II interventional trial of pediatric patients with risk factor assessment

A Phase II trial evaluating third-party VSTs for refractory viral infections in pediatric patients post-HSCT or with IEI. VSTs were expanded using a rapid expansion protocol in G-Rex-10 for 10 days. Clinical response rate was 62%.

Human 3D Ovarian Cancer Models Reveal Malignant Cell–Intrinsic and –Extrinsic Factors That Influence CAR T-cell Activity

The study uses 3D ovarian cancer models (spheroids, collagen gels, microfluidics) to study MUC1/TnMUC1 CAR T-cell activity. Resistance mechanisms and fibroblast interactions are explored. Transduced CAR T cells were cultured and expanded in G-Rex plates for 10 days.

A quality-by-design approach to improve process understanding and optimise the production and quality of CAR-T cells in automated stirred-tank bioreactors

This study uses Quality-by-Design (QbD) and Design of Experiments (DOE) to optimize CAR-T expansion. Small-scale DOE studies were conducted in G-Rex 24-well plates to identify optimal parameters (single activation, short seed train), which were then translated to Ambr 250 bioreactors.

Selection, engineering, and in vivo testing of a human leukocyte antigen–independent T-cell receptor recognizing human mesothelin

The study describes a mesothelin-specific HLA-independent TCR (HiT) that activates T cells and kills tumor cells in vitro and in vivo. Large-scale T-cell transduction and expansion were performed using G-Rex 6M plates.

T antigen–specific CD8+ T cells associate with PD-1 blockade response in virus-positive Merkel cell carcinoma

The study analyzes T-Ag-reactive CD8+ T cells in Merkel cell carcinoma patients treated with anti-PD1. Responders showed broader T-cell recognition. T-Ag-specific T cells were expanded from PBMCs using artificial Ag-presenting scaffolds in G-Rex 24 well plates to enhance tumor killing.

Human keratinocytes grown at a gas-permeable interface in vitro stratify correctly to generate engineered human epidermis

This study demonstrates that human keratinocytes cultured on a gas-permeable interface (GPI) stratify to form a mature epidermis, comparable to the air-liquid interface method. G-Rex 6 well plates were used as the GPI device for culturing the keratinocytes and inducing stratification.

A rescue approach in refractory diffuse large B-cell lymphoma with obinutuzumab-redirected cytokine-induced killer cells: a first-in-human case report

A case report detailing the treatment of refractory DLBCL using CIK cells combined with obinutuzumab. The therapy achieved a very good partial remission without severe toxicity. CIK cells were produced and expanded for 14 days in G-Rex using a blinatumomab-based protocol.

TALEN-edited allogeneic inducible dual CAR T cells enable effective targeting of solid tumors while mitigating off-tumor toxicity

The study develops allogeneic "Smart CAR T cells" using TALEN gene editing. These cells constitutively express a FAP CAR to target CAFs, which induces mesothelin CAR expression. This logic-gated approach improves solid tumor efficacy and safety. G-Rex 6M plates were used for the expansion of gene-edited T cells.

Conserved allomorphs of MR1 drive the specificity of MR1-restricted TCRs

This study investigates the specificity of the MR1-restricted TCR from clone MC.7.G5. Results show recognition is restricted to the MR1*04 allomorph and is not cancer-specific, reacting to healthy cells expressing MR1*04. G-Rex systems were used to seed and expand CD3+ T cells during the transduction process.
