Preclinical comparison of non-signaling domain in CD19 CAR T cell with interleukin-7 receptor alpha signaling domain

Three non-signaling domain designs were tested in a CD19 CAR with 4-1BB and IL-7R alpha. IgG2/CD28 lowered expression and function, CD8/CD8 gave the highest density, and IgG2.CH3/CD28 raised cytokines and persistence. CAR T cells were cultured in G-Rex plates (Wilson Wolf).
