Human Islet Viability and Function Is Maintained During High-density Shipment in Silicone Rubber Membrane Vessels

This study evaluates shipping human islets in high density using silicone rubber membrane (SRM) vessels (G-Rex). The method maintained islet viability and function comparable to low-density controls, allowing an entire clinical islet isolation to be shipped in a single vessel.

Human natural regulatory T cells subsets: Functional characterization and T cell receptor repertoire analysis

This dissertation analyzes human Treg subsets and their TCR repertoires. It demonstrates that expanding Tregs in G-Rex10 devices with rapamycin enhances expansion (at least 50-fold) compared to plates, while maintaining FoxP3 expression and suppressive function, facilitating clinical translation.

Activation and Propagation of Tumor-infiltrating Lymphocytes on Clinical-grade Designer Artificial Antigen-presenting Cells for Adoptive Immunotherapy of Melanoma

This is the published version of the study optimizing TIL propagation using K562 aAPCs. It confirms that the aAPC/G-Rex platform supports rapid expansion of TILs to clinical scales with an effector-memory phenotype, comparable to PBMC feeders. Note: This is a duplicate study of 2014_Forgetet_al_TIL.pdf.

Optimizing the production of suspension cells using the G-Rex “M” series

This study defines optimized parameters for the G-Rex 'M' series using K562 cells. It demonstrates that 1L of medium with optimal seeding allows 100-fold expansion in 10 days in the G-Rex 100M. The platform showed linear scalability from 5 to 500 cm2 and validated glucose measurement for cell quantification.

Expansion of Human and Murine Hematopoietic Stem and Progenitor Cells Ex Vivo without Genetic Modification Using MYC and Bcl-2 Fusion Proteins

This study reports the expansion of HSPCs using Tat-MYC and Tat-Bcl-2 fusion proteins. G-Rex 10 devices were used to expand CD34+ cells from cord blood, mobilized blood, and bone marrow. The expanded cells retained long-term repopulating activity and differentiated into multiple lineages in vivo.

Large-scale myeloid dendritic cells generation from umbilical CD34+ cells using G-rex

This conference paper reports a method for large-scale generation of myeloid dendritic cells (DCs) from cord blood CD34+ cells using G-Rex10. The G-Rex system yielded over 1.5 billion DCs, significantly outperforming conventional culture plates, and produced DCs capable of inducing Th1 responses.

Kinetics of Tumor Destruction by Chimeric Antigen Receptor-modified T Cells

This research investigates the kinetics of tumor destruction by CAR T cells targeting MUC1 and PSCA. G-Rex 100M devices were used to expand the CAR-modified T cells. The study found that dual targeting improved efficacy but antigen intensity influenced killing; epigenetic modulation enhanced tumor susceptibility.

Antigen Presenting Cell-Mediated Expansion of Human Umbilical Cord Blood Yields Log-Scale Expansion of Natural Killer Cells with Anti-Myeloma Activity

This study utilized G-Rex to expand NK cells from cord blood using artificial antigen presenting cells (aAPCs). The protocol achieved a mean 2389-fold expansion of high-purity NK cells in 14 days. Expanded cells demonstrated significant in vitro and in vivo cytotoxicity against multiple myeloma.

Generation of tumor antigen-specific T cell lines from pediatric patients with acute lymphoblastic leukemia – implications for immunotherapy

This study demonstrates the generation of T-cell lines targeting WT1, Survivin, MAGE-A3, and PRAME from pediatric ALL patients using autologous DCs and PHA-blasts. The cells showed specific anti-leukemic activity. G-Rex are referenced as a strategy for scaling up to clinical cell numbers.

Improved Homing to Bone Marrow, Spleen and Lung of Adoptively Infused NK Cells Expanded Ex Vivo with the Small Molecule Nicotinamide Using Feeder-Free Conditions

This study demonstrates that nicotinamide (NAM) enhances the homing and retention of NK cells in bone marrow, spleen, and lung. NK cells were expanded in G-Rex devices under feeder-free conditions with IL-2 or IL-15. The G-Rex platform supported large-scale expansion, yielding high-purity NK cells.

Safety and clinical efficacy of rapidly-generated trivirus-directed T cells as treatment for adenovirus, EBV, and CMV infections after allogeneic hematopoietic stem cell transplant

This study reports a simplified method to generate T cells targeting EBV, CMV, and Adenovirus using nucleofected dendritic cells and G-Rex devices. The 10-day G-Rex protocol produced clinically relevant cell numbers. Infused cells were safe and effective against active viral infections in HSCT recipients.
