Manufacturing CD20/CD19-targeted iCasp9 regulatable CAR-TSCM cells using a Quantum pBac-based CAR-T engineering system

This paper describes a virus-free CAR-T manufacturing process using the Quantum pBac system and Quantum Booster supplement. Use of G-Rex 24-well and 1L vessels allowed for the generation of clinically relevant numbers of high-quality CAR-TSCM cells in 10 days.

Pretreatment with IL-15 and IL-18 rescues natural killer cells from granzyme B-mediated apoptosis after cryopreservation

This study identifies that GZMB leakage causes apoptosis in cryopreserved NK cells and that IL-15/IL-18 pretreatment mitigates this. NK cells were isolated and expanded for 15-20 days in G-Rex 6-well plates using irradiated K562 feeder cells.

Bispecific CD33/CD123 targeted chimeric antigen receptor T cells for the treatment of acute myeloid leukemia

The researchers designed a split-signaling bispecific CAR to target AML cells while sparing HSCs. CAR T cells were expanded using G-REX plates to generate sufficient cell numbers for in vivo xenograft experiments comparing bispecific and monospecific CARs.

HER2 and HLA- A*02 dual CAR- T cells utilize LOH in a NOT logic gate to address on- target off- tumor toxicity

The study presents a logic-gated CAR-T strategy to safely target HER2 by sparing normal HLA-A*02+ cells. G-Rex 24 or 6-well plates were utilized for the expansion of CAR-T cells after lentiviral transduction, supporting the generation of cells for in vitro and in vivo assays.

CD8α Structural Domains Enhance GUCY2C CAR-T Cell Efficacy

This paper demonstrates that the CD8α hinge/transmembrane domain improves GUCY2C CAR-T cell affinity and antitumor efficacy compared to CD28 domains. CAR-T cells were expanded ex vivo for 12 days using G-Rex 6-well plates prior to cryopreservation.

Modular chimaeric cytokine receptors with leucine zipper enhance the antitumor activity of CAR T cells via JAK/STAT signalling

The authors developed leucine zipper-based receptors (ZipRs) to mimic cytokine signaling in CAR-T cells, enhancing their function. Autonomous cell outgrowth assays were conducted by seeding ZipR-transduced T cells in G-REX 6-well plates to ensure safety.

Silicone rubber membrane devices permit islet culture at high density without adverse effects

This study demonstrates that G-Rex devices allow for a 20-fold increase in islet seeding density compared to T-flasks without compromising viability or function. Modeling and experiments confirm that the gas-permeable membrane prevents anoxia in high-density cultures.

Sequential Exposure to IL21 and IL15 During Human Natural Killer Cell Expansion Optimizes Yield and Function

The authors found that sequential stimulation with mbIL21 then mbIL15 feeder cells optimizes NK cell yield and function. Purified NK cells were co-cultured with irradiated K562 feeders in G-Rex 6-well plates, facilitating robust expansion for functional assessment.

Rapid parallel reconstruction and specificity screening of hundreds of T cell receptors

This protocol details a medium-throughput method to clone and screen TCRs against various antigens using combinatorial dye staining. G-Rex 24-well plates are recommended for expanding TCR-transduced T cells after Dynabead removal to ensure sufficient cell numbers for screening.

Efficacy and safety of autologous tumor- infiltrating lymphocytes in recurrent or refractory ovarian cancer, colorectal cancer, and pancreatic ductal adenocarcinoma

This clinical trial assesses TIL therapy in refractory solid tumors. While no objective responses were seen, disease control was observed. The Rapid Expansion Protocol (REP) utilized G-Rex 10 and 100M to generate the large numbers of TILs required for patient infusion.

Targeting of intracellular oncoproteins with peptide-centric CARs

The study describes PC-CARs targeting the neuroblastoma oncoprotein PHOX2B presented on HLA. These CARs kill tumor cells across multiple HLA allotypes. Primary human T cells were activated and expanded in G-Rex system vessels (Wilson Wolf) during the retroviral and lentiviral transduction processes.
