Investigated methods to overcome fratricide in NKG2D-CAR T cell production. The manufacturing process included an expansion phase in G-Rex containers, which was part of the upscaling strategy to produce sufficient cell yields.

CAR T cell therapy for breast cancer: harnessing the tumor milieu to drive T cell activation
Generated MUC1-specific CAR T cells co-expressing an inverted cytokine receptor (4/7ICR) for breast cancer. Co-culture experiments to monitor anti-tumor activity were conducted in 6-well G-Rex devices using a 3D algimatrix bioscaffold.

Lym-1 Chimeric Antigen Receptor T Cells Exhibit Potent Anti-Tumor Effects against B-Cell Lymphoma
Evaluated Lym-1 CAR T cells against B-cell lymphoma. Primary T cells were transduced and expanded in 24-well G-Rex plates. The cells showed potent anti-tumor effects in vitro and in vivo against Raji xenografts.

Study on 4-1BB agonist enhancing CD8+ TIL growth from pancreatic ductal adenocarcinoma (PDAC). The Rapid Expansion Protocol (REP) was performed in G-Rex 10 devices, enabling the exponential growth of sorted CD8+ and bulk TIL populations for functional assays.

Toward a Rapid Production of Multivirus-Specific T Cells Targeting BKV, Adenovirus, CMV, and EBV from Umbilical Cord Blood
Developed a rapid method to produce multivirus-specific T cells from cord blood using overlapping PepMixes. G-Rex culture devices maintained or improved antigen recognition and overcame limitations in expanding sufficient cells for clinical application, allowing large-scale production.

Large-scale isolation and cytotoxicity of extracellular vesicles derived from activated human natural killer cells
Describes a method for large-scale isolation of extracellular vesicles (EVs) from activated NK cells. The G-Rex cell culture device allowed for the expansion of 1-3 billion aNK cells per 100 cm2, increasing EV production while reducing cost and complexity. Isolated EVs showed cytotoxicity against cancer cells.

A single exercise bout enhances the manufacture of viral-specific T-cells from healthy donors: implications for allogeneic adoptive transfer immunotherapy
This study showed that collecting PBMCs after exercise increases the yield of VSTs. The VSTs were expanded in G-Rex10 for 8 days, demonstrating that exercise is a simple adjuvant for improving VST manufacture.
